Abstract

Methylobacter alcaliphilus 20Z cells oxidized and assimilated methaneat the highest rate at pH 9.0 in the presence of 0.7 hi NaCl, i.e., under conditions optimal for growth of this culture. The cells did not assimilate methane at pH 10 in the absence of NaCl. The activity of membrane-bound NADH dehydrogenase exhibited two in vitro optima (at pH 7.8 and 8.7), was stimulated by 100 mM NaCl, and increased with the pH of the medium in which the cells had been grown. Cells grown at pH 9.0in the presence of NaCl had a higher content of cytochrome c, acid phospholipids, phosphatidylcholine, unbranched fatty acids, and some amino acids (glutamate, alanine, and aspartate) than cells grown at pH 7.0 without NaCl. Cell motility depended on Na+ and was sensitive to monensin and to the combinations DCC + HQNO and DCC + CCCP + KCN but not to arsenate or vanadate. The role of Na+ in the energetics of the methanotroph is discussed.
