Abstract

Binding of P2Y(2) purinergic agonists to their target receptor has been shown to stimulate the natural processes of mucosal hydration and mucociliaryclearance in the airways. The uridine nucleotide UTP is the first acrosolized P2Y(2) receptor agonist in clinical studies, and is being evaluated in phase II clinical trials as an acute-use agent to stimulate the expectoration of a deep-lung sputum sample via a mucociliary clearance mechanism for use in a variety of diagnostic procedures. A next-generation P2Y(2) agonist,INS365, will move into phase II clinical development as a chronic treatment for obstructive respiratory disorders, such as chronic bronchitis. These compounds represent a unique approach to disorders with a high unmet medical need, and arc expected to provide significant medical benefit and improvement in the patient's quality of life.
