Abstract

A quantitative detection of protein was achieved by a simple and inexpensive diode laser/fiber-optic colorimetric spectrometer with an optical probe with a 2 mm gap between fibers. The system consisted of a visible diode laser, a photodiode detector, and a pair of optical fibers. In this study, a droplet sample of protein, colored by the Lowry method was placed at the gap, serving as a windowless sample cell. The volume of the sample was 50 muL, which was one hundredth of the cuvette volume for a conventional UV/VIS spectrometer. Despite the micro-quantity, the dynamic range and detectability of the system were comparable to those of the conventional spectrometer. This analyzer showed the potential of microanalysis for expensive chemicals of interest.
