Abstract

Simultaneous determination of three plasma catecholamine metabolites, HVA,DOPAC, and MHPG, by means of a new sensitive high performance liquid chromatographic method with electrochemical detection of the amperometric kind, has been implemented. The chromatographic analysis was carried out isocratically on a reversed-phase column (C8 150 x 4.6 mm ID, 5 mum) with a mixtureof methanol and an aqueous solution of citric acid, octansulfonic acid, and EDTA pH 2.9 as the mobile phase. The oxidation voltage was +800 mV and the cell and column temperature was 30 degreesC. An original pretreatment of the biological samples was carried out using an SPE procedure for the extraction of three metabolites from human plasma with a high extraction yield and good selectivity. Linearity was observed in the 0.5-50 ng mL(-1) range. During validation, the repeatability (RSD%) for the spiked blank plasma formetabolite levels of 5-10-20 ng mL(-1) was found to be lower than 3.3% andthe intermediate precision lower than 3.8%. The results obtained analyzingreal plasma samples, from young volunteers under experimentally induced psychological stress, were satisfactory in terms of precision and accuracy.
