Abstract

The introduction of more intensive chemotherapy regimens has improved the prognosis of children and adults affected by acute lymphocytic leukemia (ALL). Antracyclines have become a fundamental part of ALL therapy. Data from the literature and two recent studies from the author's institution indicate that a higher dose and dose intensity of antracyclines increase the complete remission rate and the long-term outcome by reducing the occurrence of relapses. Some data suggest that the more critical period for administeringthese high doses of antracyclines is the early phase of the disease, before the development of drug resistance mechanisms. In sum, the dose and dose intensity of antracyclines given early in the disease course seem to be important factors in the improved results observed in ALL therapy.
