Abstract

The effects of permeable analogs of cAMP (Sp-5,6-DCl-cBIMPS) and cGMP (8-pCPT-cGMP)on 4-aminopyridine (4-AP) sensitive delayed rectifier (K-dr) currents of freshly isolated rat mesenteric artery smooth muscle cells (SMC) were studied using the patch-clamp technique. Application of Sp-5,6-DCl-cBIMPSat concentrations of 0.1 and 0.5 mM reduced the amplitude of K-dr currentsby 18% and 23%, respectively. Using a 3-step stimulation protocol for registration of K-dr and K-Ca currents simultaneously in individual cells, we found that application of Sp-5,6-DCl-cBIMPS reduced K-dr currents and activated K-Ca currents. Application of the permeable cGMP analog 8-pCPT-cGMP (0.1 mM) also decreased the amplitude of K-dr currents by 21%. Presence of protein kinase G (PKG) in concentration of 10 U/ml in the pipette solution abolished the effect of 8-pCPT-cGMP on the amplitude of K-dr currents. Our results suggest that cAMP and cGMP do not activate but rather weakly inhibit K-dr currents of freshly-isolated SMC of rat mesenteric artery. Possible-involvement of phosphorylation of channel proteins in the observed effects of cyclic nucleotides is discussed.
