Abstract

Antisense oligonucleotides are molecules that are able to inhibit gene expression being therefore potentially active for the treatment of viral infections or cancer. However, because of their poor stability in a biological medium and their weak intracellular penetration. colloidal drugs carriers such as liposomes were developed for the delivery of oligonucleotides. We describe in this review the different types of liposomes that were used for the delivery of oligonucleotides. Most of the studies were can-led our with cationic liposomes which form stable complexes with the negatively charged oligonucleotides. However, more sophisticated liposomal systems have been considered as pH-sensitive liposomes which can improve the endosomal escape of oligonucleotides after endocytosis, immunoliposomes which are able to deliver these molecules intracellularly through a receptor-mediated endocytosis. Plasma or lysosome membrane fusion may also be obtained through the use of fusogenic liposomes. Finally. Stealth liposomes, which are long-circulating vesicles, induce the accumulation of ODN into tumour tissues due to thehigh permeability of the endothelium in these tissues. This review discusses the advantages and disadvantages of these different strategies for the delivery of antisense oligonucleotides.
