Abstract

This paper describes a new method based on image analysis to study water uptake in tablets. To ensure a continuous water feed, the tablet is placed on a chromatographic paper band with both ends soaking in distilled water. From the side view ofthe tablet, diameter and thickness can be automatically and simultaneously measured at regular intervals. In this work, the measurements were performed on directly compressed lactose-based tablets containing Explotab, Kollidon CL, or Ac-Di-Sol as disintegrants in concentrations from 1 to 5%. Results show that the disintegrant type and its concentration modify tablet swelling, mainlyon the tablet's axial component. The derivative ofthe curve reaches amaximum in the first few seconds, and the expansion rate al 15 s (ER15) is proposed as a new parameter for the study of tablet swelling. Differences between the tested formulations were visualized through ER15, and a linear relationship was found with the concentration of the disintegrant. In conclusion, this new dilatometric technique based on image analysis appears to fit the requirements of a rapid and precise method for the study of tablet swelling.
