Abstract

By means of microelectrode techniques, the effects of acetylcholine (ACh),10(-5) M, and nibentan, 10(-7) and 10(-6) M, were studied separately and in combination on epicardial and endocardial action potential of canine right atrial free wall at cycle lengths of 250 to 2000 ms. In control action potential duration (APD) to 90% repolarization (APD(90)) was longer in endocardium at all cycle lengths. Nibentan alone induced similar concentration- and rate-dependent prolongation of APD(90) in either tissue. ACh alone shortened APD(90) in both tissues with more prominent effect in endocardium which resulted in reversal of the gradient of repolarization across the atrial wall: APD(90) in endocardium was shorter than in epicardium at all cycle lengths. Nibentan concentration-dependently antagonized ACh-induced shortening of APD(90) in endocardium but not in epicardium. As a result, at 10(-6) Mof nibentan, no difference in APD(90) between epicardium and endocardium was seen in the presence of ACh. This effect of nibentan to prevent muscarinic receptor-mediated influence on gradient of repolarization across the atrial wall may contribute to its efficacy against vagally induced atrial flutter and fibrillation.
