Abstract

Migraine pathophysiology has been much debated over many years. The recentdevelopment of relatively specific antimigraine compounds in the form of 5-HT1B/1D agonists whose clinical actions are restricted to neurovascular headaches has permitted the study of some of the fundamental mechanisms involved in the production of the pain of migraine. Rizatriptan is one of the new potent specific 5-HT1B/1D agonists whose preclinical actions include selective cranial vasoconstriction, inhibition of peripheral trigeminal afferents, and attenuation of the activity of neurons in the trigeminocervical complex. Tools characterized within the rizatriptan development program have clarified the importance of the 5-HT1B receptor for the vascular action and the 5-HT1D receptor for the neural actions of the triptans as a class.
