Abstract

Shoot tips isolated from preconditioned shoot cultures of Holostemma annulare, an endangered medicinal plant of the Indian sub-continent were successfully cryopreserved after encapsulation, dehydration and preculture in the dark in modified MS medium containing 0.5-0.75 M sucrose and 3-5% DMSO. Preculture in sucrose alone for 2-3 days was sufficient to obtain 46-56% survival and 42-52% shoot regeneration rates from the cryopreserved shoot tips. A 3-day preculture with 3 and 5% DMSO enhanced the survival rates to 72-76%and regeneration rates to 67-68% but all of the enhancement came from cells producing callus. Shoot regenerations from these shoot tips precultured with 3 and 5% DMSO occurred only at 54 and 30% rates respectively. Prolongedpreculture (> 3 days) in presence of DMSO reduced the survival of shoot tips exposed to LN.
