Abstract

Specific receptors for vasoactive intestinal peptide (VIP) exert multiple functions in the gastrointestinal tract, and have been implicated in the pathogenesis of inflammatory bowel diseases. Expression of VIP and its receptor were evaluated in guinea pig cecal tissue from normal and dextran sulfate-induced experimental ulcerative colitis animals, by immunohistochemical and in situ hybridization analyses. Nerve fibers innervating the lamina propria showed increased staining for VIP in the ulcerative colitis model. Concurrently, large foamy macrophages, present in great numbers in this region and fewer so in the tunica submucosa, showed intense VIP 1 receptor immunostaining. Smaller-sized round macrophages, populating in the tunica submucosa in greater numbers than the lamina propria, showed much weaker VIP I receptor immunoreactivity. Receptor,RNA was detected in both macrophage populations by in situ hybridization. For the foamy macrophage population, both receptor RNA and protein were strongly co-expressed, while receptor RNA predominated for the smaller-sized macrophages. An increased presence of VIP in nerve fibers at sites of cecal ulceration, and the coordinately strong expression of VIP 1 receptors on large numbers of foamy macrophages in this region gives evidence of a role for this neuropeptide and its receptor in the course of ulcerative colitis.
