Abstract

In recent years, major advances have been made in the understanding of the molecular pathogenesis of amyotrophic lateral sclerosis (ALS). The most important concept appears to be that of excitotoxic damage, which has led to first small successes in therapeutic trials. Further hopes are placed in the clinical use of trophic factors and in the development of new substances which can enhance motor neuron survival. Transgenic SOD1 mice represent an important animal model, with which new therapeutic approaches can be tested. However, the course of ALS can not be altered in a meaningful way by the drugs available now or in the near future. Therefore, palliative therapy is still the most effectivemeans of maintaining quality of life as high as possible for the patients and their relatives.
