Abstract

Blood was collected from buffalo slaughtered by halal method under hygienic condition. Serum was separated with the help of cream separator. Serum proteins were precipitated with acetone, at different pH (3.0, 5.5, 8.0 and 10.5) and temperatures (4 degrees-5 degrees C and 20 degrees-25 degrees C). The highest and lowest yields were recorded at pH 3.0 at 4 degrees-5 degrees C and 5.5/8.0 at 25 degrees-30 degrees respectively. The total viable count was highest at pH 10.5 and lowest at pH 8.0. The yeast and mould count was highest at pH 5.5 and 4 degrees-5 degrees C and lowest at pH 10.5 and 25degrees-30 degrees. Coliform and Enterobacteriaceae count was below 100/g Salmonella sp. and Staphylococcus were not present in any of the samples. Moisture and ash were lowest in pH 3.0 at 25 degrees-30 degrees C and pH 3.0at 4 degrees-5 degrees C respectively. Protein was highest at pH 3.0 at 25degrees-30 degrees C. Lipid was less than 1% in all samples. pH 3.0 at 25 degrees-30 degrees C is recommended for the large-scale production of serumprotein isolate from buffalo blood.
