Abstract

Protocorm-like bodies (PLBs) were successfully preserved without lossof viability by storage at 25 degrees C in the dark with gradual desiccation, ?This treatment also dramatically promoted leaf and root regeneration from PLBs after planting in differentiation medium, More than70% of PLBs survived and regenerated into plantlets 12 weeks after preservation under this condition, ?The best regeneration frequencies ofDarwinara cultivar Pretty Girl and Brassocattleya Pastoral 'Innocense' BM/JOGA were obtained when the PLBs were preserved for 10 weeks at 25 degrees C and 6 weeks at 25 degrees C, respectively. The relative fresh weight of the PLBs was reduced to 72% and 78%, respectively, during storage under these conditions. In contrast, PLBs preserved at 4 degrees C in the dark gradually turned brown and the regeneration frequency decreased drastically after 6 weeks of storage. When PLBs were rapidly desiccated under an air stream in a laminar flow chamber, desiccation for 30 and 120 min also promoted plantlet regeneration of both cultivars used, although the regeneration frequencies were slightly lowerthan in the gradual desiccation method.
