Abstract

The direct analysis by chiral liquid chromatography of drug enantiomers in biofluids is complicated by lack of intrinsic sensitivity due toband broadening and the problems of contaminating the chiral stationary phase. The merits of derivatization schemes based on achiral polymeric derivatization reagents for chiral drug bioanalysis are discussed. Systems based on achiral polymeric reagents designed for direct enantiomeric drug analysis are also presented. Problems with racemization and kinetic resolution are addressed, and the advantages of on-line simplicity, speed, high resolution and sensitivity are critically assessed.
