Abstract

The inhibitory property of divalents cations on mast cells secretion has been originally reported in 1957 (Zn2+) and 1971 (Mn2+). These results have been often confirmed, mast cells being stimulated by the various known triggers. The interpretation of these results has been complicated due to the poor knowledge of the Ca2+ pathways in mast cells. The variations of intracellular Ca2+ levels are prominent features of secretion processes as well as of contraction and metabolic events. However the non-excitable property of mast cells is relevant of a particular calcium homeostasis, the membrane of these cells being devoid of classical calcium channels. Thus, the potent inhibitors of mast cell secretion, Mn2+ and Zn2+, can be considered as putative antiallergic drugs modulating the intracellular Ca2+ pools. Recent studies show that Mn2+ inhibit a calcium release-activated calcium permeability leading to a decrease of intracellular calcium stores.
