Abstract

Oxytocin (OT) stimulates phosphoinositide (PI) hydrolysis and prostaglandin (PG) F-2 alpha secretion from the endometrium of cyclic pigs, but the presence of specific endometrial receptors for OT has not been demonstrated in this species. Two experiments were performed to detectthe presence of functional OT receptors on endometrium collected 15 days post estrus from cyclic gilts. OT receptor density and K-d were determined by receptor assay and Scatchard analysis. Hydrolysis of PI (i.e., incorporation of [H-3]inositol into total inositol phosphates) and PGF(2 alpha) secretion were studied with use of incubations of endometrial explants. Concentrations of PGF(2 alpha) were log-transformed for analysis of variance and are expressed as means +/- standard error of log-transformed data. In experiment 1, mean density and mean K-d ofOT receptors on endometrium of gilts were 29.2 +/- 5.54 fmol/mg protein and 1.59 +/- 0.23 nM, respectively. OT receptor density was significantly correlated with the ability of 100 nM OT to stimulate PI hydrolysis (r = 0.83, p < 0.05) and PGF(2 alpha) secretion (r = 0.87, p < 0.10), but was not highly correlated with receptor K-d (r = -0.08, p = 0.85). In contrast, OT receptor K-d was not highly correlated with OT-stimulated PI hydrolysis (r = -0.19, p = 0.68) or OT-stimulated PGF(2 alpha) secretion (r = 0.14, p = 0.86). OT-stimulated PI hydrolysis was also significantly correlated (r = 0.80, p < 0.05) with OT-stimulated PGF(2 alpha) secretion. Receptor specificity, relative to [H-3]OT binding, was 95% for OT, 104% for arginine-vasopressin (VP), 44% for lysine-VP, 55% for the selective OT antagonist L-366948, 10% for the selective VP antagonist cyclopentamethylenepropionyl(1)-Tyr(2)-Arg(8)]-VP, and less than 0.1% each for LHRH, human angiotensin I, and human angiotensin II. In experiment 2, endometrium was incubated with 0 nM peptide, 100 nM OT or 100 nM lysine-VP for determination of PI hydrolysis andPGF(2 alpha) secretion. Both OT and lysine-VP increased PI hydrolysis(p < 0.01). In contrast, PGF(2 alpha) secretion was increased by treatment with OT (p < 0.01) but not VP (p > 0.68). These results indicatethe presence of functional endometrial receptors for OT that are associated with PI hydrolysis and PGF(2 alpha) secretion during luteolysisin pigs. These results also indicate the presence of type I VP receptors that promote PI hydrolysis, but not PGF(2 alpha) secretion. Finally, lysine-VP displaced [H-3]OT binding from the endometrial membrane preparation in the receptor assay, but did not stimulate PGF(2 alpha) secretion, suggesting that lysine-VP may have an antagonistic action upon OT stimulation of PGF(2 alpha) secretion from pig endometrium.
