Abstract

Genistein, a selective protein-tyrosine kinase (PTK) inhibitor, inhibited phytohemagglutinin (PHA)-induced proliferation of equine peripheral blood mononuclear cells (PBMC) in a dose-dependent manner. The presence of exogenous interleukin-2 (IL-2) or the cyclo-oxygenase inhibitor, indomethacin enhanced the proliferation of PHA-stimulated PBMC, butfailed to reverse the immunosuppressive effects caused by genistein. Further, we found that genistein inhibited A23187 induced leukotriene B-4 (LTB(4)) synthesis. The results of this study indicate that PTK plays an important role in the signal transduction of equine PBMC, specifically for cell activation and LTB(4) synthesis.
