Abstract

In the encapsulation/dehydration technique, the key for successful cryopreservation is to induce a high level of dehydration tolerance. We succeeded in enhancing the level of dehydration tolerance using a mixture of 0.8 M sucrose plus 1 M glycerol. Cold-hardened, precultured meristems of lily with 0.3 M sucrose for 1 day were encapsulated in alginate gel beads including 0.3 M sucrose and then treated with a mixture of 0.8 M sucrose plus 1 M glycerol for 16 hr before dehydration. The encapsulated dried meristems cooled to -196 degrees C produced a very high level (about 90%) of shoot formation. This revised procedure may expand the applicability of encapsulation/dehydration technique to lessdehydration resistant materials.
