Abstract

The soil microbe, Pseudomonas fluorescens was found to grow in millimolar amounts of zinc complexed to citrate, the sole carbon source. As bacterial multiplication progressed, citrate was rapidly utilized and zinc was localized predominantly in soluble forms in the spent fluid. Dialysis experiments revealed that zinc was biotransformed from the early stages of growth. Almost 52% of zinc was complexed to moieties of molecular weight < 1 kD while the remainder was associated with protein-rich metabolites of molecular weight >3.5 kD. Atomic absorption studies indicated that the divalent metal was initially transported into the soluble cellular fraction prior to its secretion. These exocellularmetabolites appear to enable this organism to attain zinc homeostasis.
