Abstract

Paracrine regulation is now recognised to be an important control mechanism in all tissues of the body. Examples include the lymphokine system, local regulation of cell growth and the prostanoids; We have explored two endocrine paradigms - the pituitary and the islets of Langerhans. Within the pituitary we have demonstrated the local expression ofa number of neuropeptides and hypothalamic hypophyseal regulatory peptides. Each of these are localised in particular endocrine cell types and show dramatic change with alterations in the endocrine environment. For example, galanin production by the pituitary lacotroph shows at least a 2 order increase in peptide and 3 order increase in galanin mRNA when rats are treated with oestrogens. It has been well recognised that prolactin secretion is increased by oestrogens and we have been able to show that this is mediated by the local galanin. Thus immunoblockade of galanin secretion, for example in a cultured lactotroph cell line (235-1 cells), obliterates the effect of oestrogen. Investigationof paracrine regulation of the islet is at an earlier stage but this mechanism appears equally important. We have demonstrated that blockade of amylin production by the beta cell increases insulin secretion. Neuropeptide Y is locally synthesised within the islet and is greatly increased by treatment with dexamethasone. Its immunoblockade also enhances insulin release. It is still unclear to what extent paracrine regulation is physiologically important in the day to day control of endocrine function, whether its disturbance is responsible for disease, orwhether it might form a useful interventional control point for the development of new drug therapy;
