Abstract

We have previously shown that chronic infusion of interleukin-1 beta (IL-1 beta) into the brain ventricles of adult female rats caused significant and prolonged increases in progesterone (P4) secretion by the ovaries, as well as elevated prolactin (PRL) levels. The concomitant enlargement of the corpora lutea (CL), reminiscent of pseudopregnancy (PSP), suggested that the central administration of IL-1 beta over several days could prolong the life span of the CL. The hypotheses tested in the present work were aimed at determining (1) whether short-term treatment with IL-1 beta, as opposed to long-term infusion, would be sufficient to cause long-lasting increases in P4 and PRL secretion; and (2) whether the elevated plasma ACTH levels of rats injected with IL-1beta, and the subsequent increased secretion of adrenal progesterone,played a role in the ability of IL-1 beta to alter ovarian activity. Intracerebroventricular (i.c.v.) injection of IL-1 beta during estrus only, resulted in significantly elevated levels of P4 and PRL for at least 5 days, and caused persistent diestrus. The ovaries of IL-1-treated rats also contained CL of significantly increased volume compared to those of control animals. These changes were neither reversed by an anti-CRF serum (injected i.v. immediately prior to IL-1 beta) which blocked ACTH and P4 release, nor mimicked by acute exposure to mild electroshocks and the accompanying short-term increases in ACTH and P4 values. These results indicate that treatment with IL-1 beta during estrus prolongs the life span of the CL, a phenomenon accompanied by persistent increases in plasma P4 levels. The initial rise in adrenal P4 secretion caused by the stimulatory action of this cytokine on ACTH release does not appear to be necessary for this effect.
