Abstract

In the present study, the cellular distribution, tissue concentrationand molecular form of basic fibroblast growth factor (bFGF) of bovinecorpora lutea (CL) were examined at different stages of the luteal phase during the estrous cycle and pregnancy. At the cellular level (using a polyclonal antibody and the avidin-biotin immunoperoxidase technique), intense immunostaining for bFGF is exclusively observed in vascular cells during the early luteal phase (days 2-3). Luteal cells are not labelled. With increasing age of the CL, staining pattern for bFGF changes dramatically. On days 4-7 the large luteal cells exhibit a very intense and distinct labelling of the cell membrane, whereas the cytoplasm exhibit only a faint staining for bFGF. Small luteal cells are immunonegative. Vascular cells still stain positive. Beginning with the mid luteal phase (days 8-12) most of the capillary endothelial cellsare no longer positive for bFGF and the intense membrane staining of the large luteal cells is reduced. The reaction product is now localized exclusively in the cytoplasm of luteal cells. This staining patternis then characteristic for the late luteal phase, after regression and during pregnancy. Tissue concentrations (measured by specific RIA; x+/- SEM) for bFGF are highest (25.3 +/- 6.7 ng/g wet tissue) during the early luteal stage (<day 5), decrease significantly on days 6-12 (9.4 +/- 2.6) and increase during the late stage (days 14-17; 13.7 +/- 6.7 ng), after luteolysis (20.1 +/- 4.3 ng) and pregnancy (22.5 +/- 3.9ng). Western blot gives a signal at 18 kDa on day 3, faint signals at18 and 16 kDa on day 5 with an increase of the 16 kDa signal on days 14, 20 and 12 weeks of pregnancy. The results suggest that bFGF in theCL has an effect on angiogenesis during the early stage and a possible luteotropic function during the estrous cycle and pregnancy.
