Abstract

Biochemical changes occurring during the process of shoot regeneration in callus cultures of Feronia limonia (a tree plant of high medicinal value) were analysed for peroxidase, polyphenol oxidase,IAA oxidase,protein, total and orthodihydroxy phenols. While describing biochemical changes, the following stages were taken into consideration; callus, differentiating green callus, calus with distinct shoot primordia, differentiating callus with well organised multiple shoots and shoots of one year old plant which was tissue culture derived. Cotyledon tissues were used as control. It was proved that the process of dedifferentiation and redifferentiation had high metabolic activity. In differentiating callus cultures high peroxidase activity was recorded. A fifteen fold increase in polyphenol oxidase activity was observed in shoot primordia as compared to cotyledon tissues taken as control. Similarly IAA oxidase activity was also higher in differentiating shoot primordia. There was concomitant increase in total protein contents in the tissues which showed higher enzyme activities. The older and differentiated tissues showed more total phenolic contents as compared to younger tissues including control.
