Abstract

The electrochemical oxidation of 1-methyluric acid was studied at a pyrolytic graphite electrode (PGE) in the pH range 2.1-10.0. One well-defined, 2e, 2H(+), pH-dependent oxidation peak was observed in linear and cyclic sweep voltammetry. The decay of the UV-absorbing intermediate and the pseudo-first-order rate constants were calculated at different pH values and at different ionic strengths of the phosphate buffer. At pH 3.0, the electro-oxidation products were characterized as 1-methylalloxan and urea, whereas at pH 7.0, the major product of oxidation was found to be 5-hydroxyhydantoin-5-N-methylcarboxamide. A tentative EC mechanism for the oxidation has been suggested. A comparison of the electrochemical and peroxidase-catalysed oxidation of 1-methyluric acid indicates that both oxidations proceed by an identical mechanism. On the basis of these results, it is concluded that electrochemical investigations of purines when coupled with spectral studies reveal theprobable reaction routes of biological oxidation.
